Beagle Dog Treated Subcellular Fractions
View Products XenoTech provides hepatic subcellular fractions from sexually-mature, male Beagle dogs treated with prototypical inducers to serve as positive controls for ex vivo enzyme induction studies.
These products are pooled from many animals to ensure good representation of the inducer and to provide sufficient product availability. The following lot-specific product information is included with each order of microsomes.

Induction of specific cytochrome P450 enzymes are obtained through the treatment of dogs with the following prototypical inducers.
β-NaphthoflavoneTreatment of male dogs with β-naphthoflavone causes a marked induction (>10-fold) of liver microsomal CYP1A levels, which is associated with an increase in 7-ethoxyresorufin O-dealkylation, 7-methoxyresorufin O-dealkylation and benzo[a]pyrene 3-hydroxylation. Vehicle control: D1098. Dosing regimen: 10 mg/kg body weight single injection per day on days 1-4, liver is harvested and snap-frozen on day five.
PhenobarbitalTreatment of male dogs with the peroxisome proliferator, phenobarbital, causes a marked induction (>10-fold) of liver microsomal CYP2B levels, which is associated with an increase in 7-pentoxyresorufin O-dealkylation, 7-benzyloxyresorufin O-dealkylation and testosterone 16β-hydroxylation. Vehicle control: D1073. Dosing regimen: 10 mg/kg body weight single injection per day on days 1-2, 20 mg/kg body weight single injection per day on days 3-6, 30 mg/kg body weight single injection per day on days 7-14, liver is harvested and snap-frozen on day fifteen.
RifampinTreatment of male dogs with rifampin causes a marked induction (>2-fold) of liver microsomal CYP3A levels, which is associated with an increase in testosterone 6β-hydroxylation. Vehicle control: D1098. Dosing regimen: 10 mg/kg body weight single injection per day on days 1-4, liver is harvested and snap-frozen on day five.
Clofibric acidTreatment of male dogs with the peroxisome proliferator, clofibric acid, causes minimal induction (<2-fold) of liver microsomal CYP4A levels, which is associated with an increase in lauric acid 12-hydroxylation. Note: Clofibric acid is not the same as clofibrate, although both compounds are peroxisome proliferators and CYP4A inducers. Clofibrate is the ethyl ester of clofibric acid and is not readily soluble in water, in contrast to the free acid, clofibric acid. Vehicle control: D1073. Dosing regimen: 10 mg/kg body weight single injection per day on days 1-4, liver is harvested and snap-frozen on day five.
Saline (control)Liver microsomes from male dogs treated with saline are intended to serve as vehicle-treated controls for studies with liver microsomes from male dogs treated with certain P450 enzyme inducers, such as phenobarbital and clofibric acid. Dosing regimen: 5 mL/kg body weight single injection per day on days 1-4, liver is harvested and snap-frozen on day five.
Corn oil (control)Liver microsomes from male dogs treated with corn oil are intended to serve as vehicle-treated controls for studies with liver microsomes from male dogs treated with certain P450 enzyme inducers, such as β-naphthoflavone, and rifampin. Dosing regimen: 5 mL/kg body weight single injection per day on days 1-4, liver is harvested and snap-frozen on day five.
