Human CYP bDNA Probes
The performance of the bDNA probe sets was verified in cultured human hepatocytes. For example, hepatocytes from human livers HL178 and HL180 were treated for three days with 100 µM omeprazole, 10 µM rifampin or vehicle (0.1% DMSO). The level of CYP mRNA was measured and normalized to that of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA. The induction of CYP1A2 mRNA by omeprazole ranged from 19- to 25-fold, whereas CYP3A4 mRNA induction by rifampin ranged from 8- to 19-fold.
